As a result, domperidone is now recommended only for short-term use at the lowest effective dose in adults and adolescents over 12 years of age.

What Conditions Is Domperidone Used For?

How Does Domperidone Work?
Domperidone works by blocking peripheral dopamine D2 receptors in two key locations. In the stomach and upper gastrointestinal tract, it blocks the inhibitory effect of dopamine on smooth muscle, enhancing gastric motility, accelerating gastric emptying, and improving coordination between the stomach and duodenum — its prokinetic effect.
As an antiemetic, domperidone acts at the chemoreceptor trigger zone (CTZ) in the area postrema — a region at the base of the brain that sits outside the blood-brain barrier and can detect nauseating stimuli in the bloodstream. By blocking dopamine receptors at this site, domperidone suppresses the vomiting reflex without causing the central dopamine blockade associated with drugs that penetrate the brain fully.
Domperidone also raises prolactin levels by blocking dopamine's inhibitory effect on the pituitary gland — an endocrine effect that may result in galactorrhoea (breast milk production), gynaecomastia, or menstrual irregularities in some patients.

Administration Tips
Take domperidone 15 to 30 minutes before meals and, if required, before bedtime. Taking it before eating allows the drug to enhance gastric motility at the time food enters the stomach.
